Tirzepatide acts on two hormone receptors: GIP (glucose-dependent insulinotropic polypeptide) and GLP-1 (glucagon-like peptide-1). These pathways help regulate appetite, food intake and blood sugar.

How these medicines work

These medications act on hormone pathways involved in appetite and food intake. Medication may help eligible patients manage hunger as part of a plan that includes a reduced-calorie diet and increased physical activity.

Who may be eligible?

Adult weight-management eligibility commonly includes a BMI of 30 or higher, or 27 or higher with at least one weight-related condition, such as high blood pressure or high cholesterol. BMI alone does not determine suitability. Your provider reviews your medical history, medicines, risks and goals before recommending treatment.

Bring a list of medicines and supplements, previous weight-management treatments, and questions about your goals. Discuss your health conditions, allergies and pregnancy plans. If medication is appropriate, your provider will explain the exact product, administration, monitoring and follow-up. Prescription and availability depend on your assessment.

Important safety considerations

Semaglutide and tirzepatide carry boxed warnings about thyroid C-cell tumors observed in rodents; it is not known whether they cause these tumors in humans. They must not be used with a personal or family history of medullary thyroid carcinoma or MEN 2, or a serious allergy to the prescribed product.

Common side effects include nausea, vomiting, diarrhea and constipation. Serious risks can include pancreatitis, gallbladder problems and dehydration-related kidney injury. Low blood sugar risk can increase with some diabetes medicines. Discuss pregnancy, breastfeeding, severe stomach problems and planned anesthesia with your provider. Seek urgent care for severe persistent abdominal pain or a serious allergic reaction.
